Ceita,Geruza de Oliveira; Vilas-Boas,Laurival Antônio; Castilho,Marcelo Santos; Carazzolle,Marcelo Falsarella; Pirovani,Carlos Priminho; Selbach-Schnadelbach,Alessandra; Gramacho,Karina Peres; Ramos,Pablo Ivan Pereira; Barbosa,Luciana Veiga; Pereira,Gonçalo Amarante Guimarães; Góes-Neto,Aristóteles.
The phytopathogenic fungus Moniliophthora perniciosa (Stahel) Aime & Philips-Mora, causal agent of witches' broom disease of cocoa, causes countless damage to cocoa production in Brazil. Molecular studies have attempted to identify genes that play important roles in fungal survival and virulence. In this study, sequences deposited in the M. perniciosa Genome Sequencing Project database were analyzed to identify potential biological targets. For the first time, the ergosterol biosynthetic pathway in M. perniciosa was studied and the lanosterol 14α-demethylase gene (ERG11) that encodes the main enzyme of this pathway and is a target for fungicides was cloned, characterized molecularly and its phylogeny analyzed. ERG11 genomic DNA and cDNA were...

Mycophagous Drosophilidae (Diptera) guild and their hosts in the Brazilian Amazon Pap. Avulsos de Zool. (São Paulo)
Santa-Brígida,Rosângela; Wartchow,Felipe; Medeiros,Priscila Sanjuan; Gottschalk,Marco Silva; Martins,Marlúcia Bonifácio; Carvalho,Claudio José Barros de.
Abstract The composition and potential hosts of mycophagous Drosophilidae from a section of the Brazilian Amazon forest in the Caxiuanã National forest were investigated. Sampling was performed in three different periods at long the wet season (January (beginning) and July (end) 2013 and May (middle) 2014). The samples were collected from existing trails by actively searching for fungal fruiting bodies where Drosophilidae were present. We present composition and richness analysis over two years of sampling sampling Drosophilidae and Fungi. We evaluate sampling completeness using asymptotic species richness estimators. Out of 159 fruiting body samples and 64 fungal species, 5,124 drosophilids belonging to 55 species and 5 genera were collected. The...

Phylogenetic placement of Tritirachium strains from the URM culture collection originally founded by Augusto Chaves Batista (1916-1967) in Brazil, and the description of T. batistae sp. nov. Acta Botanica
Bezerra,Jadson Diogo Pereira; Felipe,Maria Tamara de Caldas; Paiva,Laura Mesquita; Magalhães,Oliane Maria Correia; Silva-Nogueira,Eliane Barbosa da; Silva,Gladstone Alves da; Souza-Motta,Cristina Maria de.
ABSTRACT Twenty-seven Tritirachium strains were present in the URM culture collection originally founded in Brazil by Augusto Chaves Batista. Fifteen freshly-prepared cultures were obtained from these original strains preserved under mineral oil. DNA was extracted for analysing phylogenetic relationships using the sequence information available from Tritirachium type materials and reference strains. Phylogenetic analysis using the internal transcribed spacer (ITS) ribosomal DNA sequences revealed that eight of these strains belong to the same clade as of T. oryzae and that the four other strains belong to the same clade as of T. candoliense. The strain URM 38, which was previously identified as “T. brumptii”, appeared to exist as a single lineage, related...

Taxonomic studies of Amanita muscaria (L.) Lam (Amanitaceae, Agaricomycetes) and its infraspecific taxa in Brazil Acta Botanica
Wartchow,Felipe; Maia,Leonor Costa; Cavalcanti,Maria Auxiliadora de Queiroz.
We analyzed specimens identified as Amanita muscaria, some recently collected and others already deposited in herbaria, in Brazil. We concluded that two subspecies of A. muscaria occur in Brazil: A. muscaria var. muscaria; and A.muscaria var. flavivolvata. The first taxon was found in association with Castanea sativa, and the second (one specimen only) was found in association with Pinus and Eucalyptus spp. Morphologically, A. muscaria var. flavivolvata is distinguished by a shallower subhymenium and by basidiospores that are more elongated than are those of A. muscaria var. muscaria, which is the more widely known subspecies. We present descriptions, discussions, illustrations and a dichotomous key for these two subspecies.

Taxonomy and evolutionary relationships within species of section Rimosae (Inocybe) based on ITS, LSU and mtSSU sequence data Naturalis
Larsson, E.; Ryberg, M.; Moreau, P.-A.; Delcuse Mathiesen, Å.; Jacobsson, S..
The present study aimed at elucidating the structure of Inocybe subg. Inosperma sect. Rimosae but included also representatives from subg. Mallocybe and the genus Auritella. Phylogenetic relationships were inferred using ITS, LSU and mtSSU sequence data. The analyses recovered the ingroup as a monophyletic, strongly supported clade. The results indicate that recognizing Auritella on the genus level renders Inocybe paraphyletic. The species traditionally placed in sect. Rimosae were found to be distributed over two strongly supported clades, Maculata and Rimosae s.s. The Maculata clade clusters with sect. Cervicolores and the two represent subg. Inosperma in a strict sense. Rimosae s.s. emerges as an independent, supported clade well separated from...

The largest type study of Agaricales species to date: bringing identification and nomenclature of Phlegmacium (Cortinarius) into the DNA era Naturalis
Liimatainen, K.; Niskanen, T.; Dima, B.; Kytövuori, I.; Ammirati, J.F.; Frøslev, T.G..
Cortinarius is a species-rich and morphologically challenging genus with a cosmopolitan distribution. Many names have not been used consistently and in some instances the same species has been described two or more times under separate names. This study focuses on subg. Phlegmacium as traditionally defined and includes species from boreal and temperate areas of the northern hemisphere. Our goals for this project were to: i) study type material to determine which species already have been described; ii) stabilize the use of Friesian and other older names by choosing a neo- or epitype; iii) describe new species that were discovered during the process of studying specimens; and iv) establish an accurate ITS barcoding database for Phlegmacium species. A total...

Total lipid and fatty acid accumulation during basidiospore formation in the ectomycorrhizal fungus Pisolithus sp. Rev. Bras. Ciênc. Solo
Campos,André Narvaes da Rocha; Costa,Maurício Dutra; Tótola,Marcos Rogério; Borges,Arnaldo Chaer.
The basidiospores of Pisolithus sp. contain large amounts of lipids, indicating provision for future germination in the host rhizosphere. However, the accumulation, composition, and mobilization of lipids during formation and germination of these spores are largely unknown. In this study, lipid storage and fatty acid composition during basidiosporogenesis were analyzed in fresh basidiocarps using bright-field microscopy and gas chromatography. Abundant lipid bodies are found in the hyphae, basidia, and basidiospores of fungal basidiocarps. This evidences a considerable C transport in the basidiocarp to meet the C demand during basidiospore formation. Fatty acid composition analysis revealed the presence of 24 compounds with chains of 9 to 18 C atoms,...
